Abstract
Coronary artery disease (CAD), leading to death mostly in developed countries, is a complex genetic disorder. There is a relationship between coronary artery disease and candidate genes. Among them, matrix metalloproteinase (MMP-9) and endothelial nitric oxide syntase (eNOS) are the most studied genes. It is suggested that MMP9 -1562 C/T polymorphism might affect both gene transcription and product. The other candidate gene region is nitric oxide syntase that associated with pathogenesis of atherosclerosis. The eNOS T-786C polymorphism is essential for transcription regulation. The aim of this study is to determine the gene frequency of MMP9 -1562 C/T and eNOS T-786C polymorphisms in coronary artery disease in Turkish population. 71 patients and 78 healthy subjects were enrolled in our study. DNA was isolated from whole blood sample and genotyping was performed PCR-RFLP method. In this study, CC, CT and TT genotype frequency of MMP-9 – 1562 C/T polymorphisms was 70.4 %, 28.2 %, 1.4% in CAD patients and 73,1 %, 23,1 %, 3,8 % in control groups, respectively. In addition, TT, TC and CC genotype frequency of eNOS T-786C polymorphisms was 47.9 %, 50.7 %, 1.4 % in CAD patients and 61.5 %, 25.6 %, 12.8 % in control groups, respectively.
