Abstract
SOX14 gene is a member of the B2 subfamily of SOX genes, coding a protein with moderate transcriptional repression characteristics. As well as other SOX genes within B subfamily, SOX14 gene is important for early neurogenesis. Sox14 expression is restricted to a limited population of neurons in the developing brain and spinal cord of chicken and mouse. This gene marks a subset of interneurons at a defined dorsoventral position adjacent to ventral motor neurons in the spinal cord. Sox14 is also involved in other developmental processes such as in outgrowth of the apical ectodermal ridge of developing limb bud. Previously, it has been shown that a 1.3kb fragment of the SOX14 5' noncoding sequence has promoter activity in transiently transfected HepG2 cells. We further analysed this fragment in order to map positive and negative control elements within SOX14 promoter. We have cloned truncated fragments in pBLCAT5 vector in front of the tk promoter driving cat reporter gene. Fragment -790/-470 had the ability to notably enhance the activity of tk promoter. Within this region Matinspector analysis revealed numerous potential binding sites for various transcription factors, such as: TGIF, TCF/LEF1, AREB6, Foxa2, MEIS1 and MAZF. Further analyses will show which transcription factors are involved in this enhancer activity.
