PP111. POLYMORPHISM OF CANDIDATES CARDIOVASCULAR DISEASES GENES IN TATARS WITH AGE GRADATION

Abstract

The aim of the present study was to investigate the molecular-genetic basis of lifespan in Tatars from Bashkortostan. The analysis of genotypes and allele frequency distribution of angiotensin-converting enzyme gene (ACE, I/D), apolipoprotein E gene (APOE, Cys112 → Arg, Arg158 → Cys) and paraoxonase gene (PON1, Q192R) polymorphisms in connection with age gradation, using PCR and PCR-RFLP methods was performed. We have examined 1116 persons, which were differentiated on separate groups according to age gradation: the young group (n=163, 1-20 years), maturity (n=239, 21-55 years), elderly (n=356, 56-74 years), senile (n=253, 75-89 years) and long-livers (n=105, 90-108 years). It was established that in senile group, in comparison with maturity group, genotype ACE*I/*D frequency was lower (P=0.008) and genotype ACE*I/*I frequency was higher (P=0.04). In senile and elderly groups, in comparison with young group, genotype PON1*Q/*Q frequency was higher (P=0.02). Genotype PON1*R/*R frequency in senile group was lower than in young group (P=0.04). Group of long-livers tended to have 2.5 times higher genotype PON1*R/*R frequency than the senile group (P=0.01). Concerning APOE gene, we have not revealed statistically significant differences in comparison groups. Thus, the changes in polymorphic markers of ACE and PON1 genotypes frequencies in various age groups in Tatars (Bashkortostan) are traced: among senile the number of ACE*I/*D and PON1*R/*R genotype carriers are reduced in comparison with persons of younger age. Besides, we found the increase of genotype PON1*R/*R and allele PON1*R frequency in long-livers.
